Abstract:
PURPOSE: To obtain a new substd. 16-methyl-steroid deriv. of pregna-1,4- diene-3,20-dione useful as an intermediate for the synthesis of 16-methylene steroid.
CONSTITUTION: The substd. 16-methyl-steroid deriv. is a compd. of formula I [where a partial structure of formula II is a 3-keto-Δ4-system, a 3-keto-Δ1,4- system, etc.; R is methyl, -CH2OH, etc.; R1 and R2' are methyl, ≤10C aryl, etc.; (n) and (m) are 0 or 1; and R2 and R3 are H or R2 is F and R3 is formyloxy or acetyloxy], e.g. 21-acetoxy-16α-[bis(phenylthio)methyl]pregna-1,4,9(11)- triene-3,20-dione. The compd. is obtd. by allowing a compd. of formula III to react with a reagent of the formula R1S-CH2-NO2 in a basic medium and allowing a thiol or thiphenol of the formula HS-R1' to react on the resultant compd. of formula IV in the presence of an acid, preferably acetic acid or phosphoric acid.
