PURPOSE: To provide novel naphthalene compounds which are strong regulating substances of the secretion of insulin and useful in treating diabetes mellitus, insulin sensitivity, cardio-vascular disorders, insulinoma, etc.
CONSTITUTION: Novel naphthalene compounds are represented by formula I (wherein R is OH, alkoxy or the like; R1, R'1 and R2 are each H or alkyl; R3 and R4 are each H or R3 and R4 together form a methylene or ethylene bridge; R5 and R6 are each H, OH, acyloxy or the like; R7 and R8 are each H or alkyl; and R9, R10, R'a and R'b are each H, OH, alkoxy or the like) and include, e.g., 1,2,3,4,4αβ,5,6,7,8,8αβ-decahydro-2,3,5,6,7,8-hexahydroxy-1,4- ethanonaphthalene. Further, the compounds of formula I can be obtained, e.g. by reacting a quinone of formula II with a diene of formula III in a non-polar solvent under heating in an inert atmosphere according to the Diels-Adler reaction to form a fused ring, then reducing the resulting compound, converting the reduced compound to an epoxy derivative with the use of an oxidizing agent and hydrogenating the epoxy derivative.
