NEW MATERIAL:A plasmid having a DNA fragment containing β-tyrosinase structural gene, DNA fragment containing promoter DNA relating to manifestation of this structural gene and a gene controlling this promoter, a mini-FDNA fragment containing gene governing propagation-controlling and dividing system derived from F-factor plasmid and a gene governing automatic propagation potency derived from ColE1-system plasmid pBR322.
USE: Used for production of β-tyrosinase synthesizing L-tyrosine from phenol and L-serine or pyruvic acid and ammonia.
PREPARATION: For instance, F-factor plasmid is extracted from Escherichia coli K-12 strain, etc., by authentic method and mini-F fragment is cut out with restriction enzyme EcoRI, then the fragment is mixed with plasmid pBR322. T4DNA ligase is affected and further cleaved with restriction enzyme BamHI and then bound with β-tyrosinase gene to afford the objective plasmid pMTD1.
