Abstract:

PURPOSE: To provide a nucleic acid amplifier which can continuously effect the extraction of nucleic acids from the biological sample and the amplification reaction of the nucleic acids, thus enables efficient amplification of nucleic acids from an extremely trace amount of a sample with reduced loss of the sample.

CONSTITUTION: The amplification unit for amplifying an extremely trace amount of biological sample is equipped with the capillary reactor 4 for extraction and amplification of nucleic acids, the heat block 5 for keeping the capillary at a certain temperature, the filter 6 for holding the microparticles to which the nucleic acids are immobilized in the capillary, the capillary connectors 7, 8, the tank for cleaning solution 9, the PCR reactor 10, the nucleic acids elution tank 11, the mechanically driven pump for sending the solution 12, Switches for switching the solution flows 13, 14, 16, the flowing paths 15a to 15e, the collector for amplified sample 17, and the waste tank 18, while the analyzer unit 2 is equipped with the switch for changing the solution flows 19, the buffer solution pump 20, the capillary for electrophoresis of the nucleic acids 21, the buffer solution tank 22, the waste tank 23, the power source 36, the optical detector, and the buffer solution flow paths 28a, b, and they are controlled by the controlling unit 3 to effect continuous amplification of nucleic acids.
