Abstract:

To provide an improved identification method and production method of nucleic acid ligands based on SELEX method, nucleic acid ligands to HIV-RT, HIV-1 Rev, HIV-1 tat, thrombin and basic fibroblast growth factor proteins, and the like.

The production method of nucleic acid ligands is an identification method of a nucleic acid ligand to thrombin and comprises obtaining a nucleic acid mixture in which a sequence having a relatively high affinity to the protein is condensed by (a) preparing a candidate compound of RNA nucleic acid, (b) contacting the candidate compound with thrombin, wherein a nucleic acid having an increased affinity for thrombin may be partitioned from the remainder of the candidate mixture, (c) partitioning the members of the candidate mixture based on the affinity for thrombin, and (d) amplifying the molecule selected from the candidate mixture having relatively high affinity to thrombin.
