To provide a hybridization substrate for efficiently reducing a background noise and having a DNA chain fixed to a surface of the substrate and a complementarity test method using the substrate.
The hybridization substrate has at least one spot of probe nucleic acid fixed to the surface of the substrate. The surface of the substrate is covered with a nucleic acid monomolecular film at least near the spot. A method for manufacturing the substrate includes a process for fixing at least one spot of probe nucleic acid to the surface of the substrate and a process for rinsing the surface of the substrate by using a liquid containing the nucleic acid chain having a group for reacting to the surface of the substrate on an end. The invention includes a method for bringing a target nucleic acid into contact with the surface of the substrate on which the probe nucleic acid spot is fixed and testing complementarity of the target nucleic acid and the probe nucleic acid.
