PURPOSE: To obtain a nucleic acid compound having an orientation fixed by a cyclic skeleton, comprising a non-nucleic acid cyclic skeleton containing two functional groups in the same direction and two nucleic acid chains bonded to the skeleton, excellent in permeability of a cell membrane, useful for gene therapy by triple chain formation.
CONSTITUTION: This nucleic acid compound has an orientation fixed by a cyclic skeleton and comprises a non-nucleic acid cyclic skeleton of the formula containing two functional groups substantially in the same direction and two nucleic acid chains bonded through the functional groups to the non-nucleic acid cyclic skeleton. The compound is excellent in permeability of a cell membrane and high endocytosis action, is capable of forming triple chain with a third nucleic acid chain, high protein manifestation inhibiting efficiency and is useful as a medicine and a diagnostic for inhibiting manifestation of an etiologic gene as a application of anti-sense nucleic acid method. The compound is obtained by adding nucleic acid chains to the functional groups of a non- nucleic acid compound having the orientation fixed by the cyclic skeleton, containing two functional groups substantially in the same direction in which a distance between the two functional groups is 0.6-1.4nm.
