Abstract:

PROBLEM TO BE SOLVED: To obtain a new nucleic acid monomer comprising a specific polyfunctional nucleic acid monomer, and useful for contamination of a selective cleaving site and an abasic site to an oligonucleotide chain, and determination of the oligonucleotide, and further useful as a phosphorylation agent, etc.
SOLUTION: This nucleic acid monomer is a new polyfunctional nucleic acid monomer represented by formula I {R1 is a base-stable and acid-sensitive blocking group; R2 is a phosphoric acid derivative enabling a nucleic acid to be added to 5'-position of an oligonucleotide chain in the chemical synthesis; R3 is a halogen, methyl, I, Br or F; R4 is H car methyl; R5 is revulinyl, a group of formula II [Rk is an amino, nitro, a halogeno, OH, a lower alkyl or a lower alkoxy; (k) is 0-4], etc.; Z is a group of formula III [(x) is 1-8], etc.}. The nucleotide reagent is useful for contamination of a selective cleaving site and/or an abasic site to an oligonucleotide chain, a chemical assay such as determination of the oligonucleotide, a reagent for phosphorylation, etc.
