Abstract:

PURPOSE: To accurately detect a transformant of a vegetable by preparing an oligonucleotide having a gene coded to T-DNA of Ri plasmid of agrobacterium rhizogenes A4 strain and applying the oligonucleotide to a vegetable.

CONSTITUTION: A double-stranded nucleotide sequence is separated into single- stranded chain and the complementary chain is functioned as a template of the chain-extension reaction by the other primer. The simultaneous chain- extension reaction with two primers and the separation of the chain-extension reaction product from the template are repeated to amplify the number of copied of a specified nucleotide fragment. The amplified nucleotide fragments are detected e.g. by electrophoresis. The objective transformant can be detected by detecting the presence of a nucleotide sequence to be recognized in the vegetable specimen by the determination of the molecular weight of the fragment and/or by the nucleic acid staining process.
