Abstract:
PURPOSE: To easily prepare p-hydroxymandel compound in a high selectivity, by condensing a hydroxylated aromatic compound and a carbonyl compound using quaternary ammonium hydroxide.
CONSTITUTION: In the presence of quaternary ammonium hydroxide, a hydroxylated aromatic compound of formula I (wherein A is an optionally substituted, monocyclic- or polycyclic aromatic carbon ring residue having at least five atoms in the ring; and A has at least one OH group and the para- position of OH is unsubstituted) (e.g. guaiacol or the like) and a carbonyl compound of formula II (wherein R1 is H, 1-6C alkyl or phenyl; and R2 is an electrophilic group) (e.g. glyoxylic acid or the like) are condensed to obtain an optionally substituted p-hydroxymandel compound (e.g. 3-methoxy-p- hydroxymandelic acid or the like). Here, the quaternary ammonium hydroxide is tetramethyl ammonium hydroxide or the like. As the compound of formula I, a compound of formula III (wherein the p-position is unsubstituted; (x) is from 1 to 4; and R is H, alkyl, alkoxy or the like) is especially preferable.
