Abstract:
PURPOSE: To prepare a medicament for treating and preventing physiological disorder associated with β-amyloid peptide such as Alzheimer's disease and Down's syndrome. CONSTITUTION: This medicament contains an effective amount of vacuolar adenotriphosphatase inhibitor or a pharmaceutically acceptable salt or prodrug thereof. The compound is exemplified by a compound of formula I Y is formula II (W<1> is H or a 2-6C alkanoyl) or CH2 ; Z is formula III [W<2> is H, OH or a 1-6C alkoxy; W<3> is a 1-6C alkyl, a 2-4C alkyl, W<4> is OH, a 1-6C alkoxy, a 2-6C alkanoyl, formula IV (W<5> is OH, a 1-6C alkoxy, NH2 , formula V, etc.), etc.] or formula VI; R<1> is H, a 2-6C alkanoyloxy; R<2> is a 1-6C alkyl}, concanamycin A of formula VII, elaiophylin of formula VIII and bafilomycins.
