Abstract:
PROBLEM TO BE SOLVED: To obtain a new piperidine derivative useful as a n intermediate for medicines such as paroxetine useful as an antidepressant drug.
SOLUTION: This piperidine derivative shown by formula I (R1 is H, benzyloxycarbonyl, or t-buthoxycarbonyl; R2 is hydroxymethyl, an alkylsulfonyloxymethyl, phenylsulfonyloxymethyl, and so on; Z is ethylene or carbonyl), e.g. (3S,4R)-trans-1-benzyloxycarbonyl-4-(4-fluorophenyl)-3-[(3,4- methylenedioxyphenyl)oxymethyl]piperidine, especially a compound of formula II (R3 is benzyl or t-butyl) which is obtained by reacting a compound of formula III with a compound of the formula R3OC(=O)Y(Y is Cl and so on) to give a compound of formula IV, which is then reacted with a compound of the formula R4S(=O)2Cl (R4 is an alkyl and so on) to give a compound of formula V, which, in turn, is reacted with 3,4-methylenedioxyphenol under basic condition.
