Abstract:
PROBLEM TO BE SOLVED: To produce a polypeptide containing IgG Fc region, the polypeptide having a higher anti-inflammatory activity and a lower cytotoxicity compared to an unpurified antibody.SOLUTION: IgG sialylation is shown to be regulated by the induction of an antigen-specific immune response, and the cytotoxic and anti-inflammatory response of the IgG Fc domain are shown to result from the different sialylation of the Fc-linked core polysaccharide. The cytotoxicity of the IgG antibody is reduced by sialylation; on the contrary the anti-inflammatory activity is enhanced. The polypeptide has high sialic acid content compared to the unpurified antibody, including the Fc region of human IgG1, IgG2, IgG3, or IgG4.
