PURPOSE: To provide the subject substance having excellent solubility and used as a hemorrhage remedy for hemophilia A patients, etc., by subjecting an aqueous blood coagulation VIII factor-containing solution to a gel filtration using a water-insoluble porous gel having a specific exclusion limited mol.wt. to separate the blood coagulation VIII factor.
CONSTITUTION: The aqueous solution of the blood coagulation factor VIII is prepared by the following processes: extracting and dissolving cryoprecitate prepared by freezing and melting normal human plasma with and in a 20mM tris - 10mM citric acid buffer solution (pH7.0), continuously adding aluminum hydroxide and bentonite to the solution, stirring, centrifuging the solution to collect the supernatant, adding glycine to the supernatant, centrifuging the supernatant, adding NaCl to the treated supernatant to salt up precipitates and subsequently dissolving the salted-up precipitates in a buffer solution. The prepared aqueous solution of the blood coagulation factor VIII is subjected to a gel filtration using a water-insoluble porous gel having an exclusion limited mol.wt. of 800000-100000000 to provide the blood coagulation factor VIII.
