PURPOSE: To obtain easily the titled compound useful as a drug for adnerocortical hormone in high yield without requiring a specific solvent, by reacting 17α, 21-cyclic orthoester compound of 20-oxopregnane steroid with xanthene-9-carbonyl chloride.
CONSTITUTION: A 17α,21-cyclic orthoester compound of 17α,21-dihydroxy-20- oxopregnane sterod shown by the formulaI(the bond between 1W2 positions of the ring A of steroid nucleus is a single bond, or double bond; X1 is H, CH3, or halogen; X2 is H, or halogen; Y is oxo, OH in β coordination or halogen; Z is alkyl, alkoxy, etc. in α or β coordination; R is alkyl, aralkyl, aryl; R' is alkyl) is reacted with xanthene-9-carbonyl chloride in a solvent such as a (halogenated) hydrocarbon, tertiary amide solvent, etc. at 70W120°C, to give the desired compound shown by the formula II.
USE: Useful as remedying various kinds of skin diseases, allergic inflammation, local inflammation, etc. and as an intermediate for drugs.
