Abstract:
PURPOSE: To provide the compounds in high yields by adding an acid to a (6RS)-diasteroisomeric mixture of an alkali metal salt of 5,6,7,8-tetrahydrofolic acid and selectively crystallizing.
CONSTITUTION: By adding a small amount of an acid (e.g. hydrochloric acid) to an aqueous solution of a (6RS)-diasteroisomeric mixture of an alkali metal salt of 5,6,7,8-tetrahydrofolic acid so as to be stabilized in pH of 4.8 to 5.3 and selectively crystallizing at 5°C to 80°C, preferably at 45°C, and separating the crystals by filtration or centrifugation, (6S)-5,6,7,8-tetrahydrofolic acid of formula, diastereoisomeric purity of which is at least 75%, which enhances antitumor effect or reducing side effect when administered with a carcinostatic agent, is obtained. Relating to the process free acids which are produced as by-products are not generated because acid addition salts and not used and thereby their treatment is in necessitated, the process is economical and has no bad influence on environment.
