Abstract:

PROBLEM TO BE SOLVED: To industrially advantageously obtain the subject compound useful as an intermediate for prostaglandins simply and efficiently by reacting a specific propyne dianion with a specific ketone and quenching the reaction product in an acidic aqueous solution.
SOLUTION: Propyne is reacted with an alkyl metal of the formula R3M [R3 is a lower alkyl; M is lithium or MgX (X is a halogen)] (e.g. n butyllithium) in a solvent (e.g. tetrahydrofuran) at -100 to 40°C for one minute to six hours to give a propyne dianion of formula I. The prepared compound is reacted with a ketone of formula II [R1 and R2 are each (O-containing) 1-10C straight- chain or branched chain (unsaturated) saturated hydrocarbon, a (substituent group containing) 3-8C cycloalkyl, etc.] at -78 to 30°C for one minute to six hours and quenched in an acidic aqueous solution (e.g. a saturated aqueous solution of ammonium chloride) to give a 4-hydroxy-1-alkyne of formula III.
