Abstract:

PURPOSE: To obtain a 4-mercaptopyrrolidine compound useful as a synthetic intermediate for carbapenem-based antimicrobial agents, etc., in high yield at a low cost by reacting a specific halogenobutyric acid compound (salt) with an amine compound (salt) and, as necessary, thiocarbonylating the resultant compound and/or removing an SH-protecting group.
CONSTITUTION: (A) A compound of formula I (R11 is an SH-protecting group; R2 is H or a carboxyl-protecting group; R3 is a halogen) or its salt is reacted with (B) a compound of the formula RNH2 (R is H, a lower alkyl or a lower alkanoyl) or its salt (in a solvent, e.g. methanol at 10-100°C temperature). The resultant compound, as necessary, is thiocarbonylated (e.g. by reacting with a thioketonizing agent such as phosphorus pentasulfide in a solvent, e.g. dimethoxyethane at 10-200°C temperature) and/or the SH-protecting group is removed to afford the objective compound of formula II (R1 is H or the SH- protecting group; X is O or S).
