Abstract:
To obtain the subject compound useful as an intermediate for synthesizing compounds having insulin resistance-improving effect, by reaction of a benzylidene-thiazolidine-2,4-dione with a metal hydride.
This new compound, a 5-benzylthiazolidine-2,4-dione derivative of formula II, is obtained in high yield by reaction of a compound of formula I [R is a (substituted) arylheterocycle, heteroaromatic ring or the like; X is an amino-protecting group] with 1-10 equivalents, based on the compound of formula I, of a metal hydride (e.g. sodium borohydride) in a mixed solvent of an amide- or ether-based organic solvent (e.g. dimethylformamide) and water or an alcohol-based organic solvent (e.g. ethanol) at 0-80°C.
