To obtain the subject antibiotic useful for an antitumor agent, etc., by separating a-product as a solution from a culture product prepared by culturing a bacterium capable of producing DC-12, subjecting the solution to cyanidation treatment and extracting a reaction product with an organic solvent.
A bacterium [e.g. Streptomyces melanovinaceus DO-52 strain (FERN BP-5,911), etc.], capable of producing DC-52 of formula I is cultured to give a culture product, from which DC-52 is separated as a solution in water, a hydrous organic solvent or an organic solvent. The obtained solution of DC-52 is subjected to cyanidation treatment and DX-52-1 of formula II contained in the reaction solution is extracted with an organic solvent to efficiently give the objective antitumor antibiotic DX-52-1 of formula II. A culture supernatant liquid is prepared from the culture product obtained by culturing the bacterium capable of producing DC-52 and DC-52 can be separated by using a nonionic porous resin.
