PURPOSE: To remarkably improve reaction rate and yield, by reacting cyclodextrin with glucose or maltooligosaccharide having fluorine atoms linked to carbon atoms at the reducing ends thereof in the presence of pullulanase having specific properties.
CONSTITUTION: Cyclodextrin is reacted with glucose or maltooligosaccharide having fluorine atoms linked to carbon atoms at the reducing ends thereof in the presence of pullulanase having the following properties of afford the aimed cyclodextrin having the glucose or maltooligosaccharide linked in the form of branches. Properties of the pullulanase; (1) Produced by Aerobacter aerogenes outside the microbial cells. (2) optimum pH; 6.0W6.5 and stable range; 5.0W11.5pH. (3) optimum action temperature; 50°C and thermal stability; up to 50°C. (4) Uninhibited by Mn2+ and Ag+ with little inhibition by SH reagent. (5) Activated by Ca2+. (6) About 70,000mol.wt. without containing cysteine.
