Abstract:
PURPOSE: To obtain a heterocyclic compound in which only a hydroxyl group is protected, under a mild condition without uselessly using a dicarbonate.
CONSTITUTION: A compound of formula I (X1 is NH, S or O; X2 is CH or N; R1 is an organic group) such as ethyl 2-(2-amino-4-thiazolyl)-2- hydroxyiminoacetate is reacted with a dicarbonate of formula II (R2 is an alkyl, an alkenyl or an aralkyl) in a solvent (e.g. acetone, methanol or N,N- dimethylformamide) having ≥10 relative dielectric constant in the presence of an amine of formula III (R3 and R4 each is an alkyl or aryl or R3 and R4 form a ring) or an inorganic salt to give a compound of formula IV such as ethyl 2-(2-amino-4-thiazolyl)-2-t-butoxycarbonyloxyiminoacetate. 4-N,N- Dimethylaminopyridine or 4-piperidinopyridine is preferable as the amine of formula III.
