Abstract:

PROBLEM TO BE SOLVED: To obtain an N-acetylcyclohexylglycine compound useful as a synthetic intermediate for an oxazole-type heterocyclic aromatic compound having antipyretic action, etc., in high selectivity and yield by hydrogenating the nucleus of a specific N-acetylphenylglycine.
SOLUTION: The objective compound of formula II useful as an important synthetic intermediate for an oxazole-type heterocyclic aromatic compound having antipyretic actions, etc., especially selective inhibitory action on cyclooxygenase-2 is produced by the nucleus-hydrogenation of an N- acetylphenylglycine of formula I (R1 is H or an amino-protecting group; and R2 is H or a carboxyl-protecting group) in the presence of a nickel catalyst. The nickel catalyst is preferably Raney nickel and the amount of the Raney nickel is preferably 0.0001-0.3 pt.wt. based on 1 pt. wt. of the N- acetylphenylglycine. Water containing preferably ≥1 equivalent of a base is used as a reaction solvent. The reaction is preferably carried out at 100-210°C under a hydrogen pressure of 10-200 kg/cm2.
