Abstract:

PURPOSE: To readily and inexpensively obtain an N-guanidinothiourea salt which is useful as an intermediate for medicines and agrochemicals by carrying out the thermal reaction of hydrazine and cyanamide which are readily available in industry.
CONSTITUTION: (A) Hydrazine and (B) cyanamide are allowed to react with each other in the presence of an acid such as hydrochloric acid or sulfuric acid in a polar solvent such as water, methanol or ethanol, as they are heated, usually at 50°C - under reflux. (C) The aminoguanidine formed is heated together with (D) a thiocyanate salt in the presence of an acid, preferably at 80-120°C to give an N-guanidinothiourea salt. Further, this N-guanidinothiourea salt is heated under alkaline conditions to give this 3-amino-5-mercapto-1,2,4-triazole.
