PURPOSE: To obtain a compound useful as antiulcer agent using inexpensive phosphorus trichloride in simple treatment and good yield, by reacting an anthranilic acid lower alkyl ester with phosphorus trichloride and then reacting the resultant product with trans-4-guanidinomethylcyclohexane carboxylic acid (GCA).hydrochloride.
CONSTITUTION: An anthranilic acid lower alkyl ester is reacted with phosphorus trichloride in a solvent such as pyridine at 0°CW ambient temperature to afford a reactive derivative of anthranilic acid lower alkyl ester, which is then reacted with trans-4-guanidinomethylcyclohexane carboxylic acid (GCA).hydrochloride at ambient temperature W80°C to provide the aimed product expressed by the formula (R is lower alkyl). Phosphorus trichloride is used at an amount of 0.5W0.7mol. based on 1mol. anthranilic acid lower ester and GCA.hydrochloride is used at an amount of 0.8W1.2mol. based on 1mol. reactive derivative of anthranilic acid lower ester.
