Abstract:

PURPOSE: To obtain an allyl alcoholic derivative useful as a raw material for producing an optically active γ-lactone derivative to be a key intermediate for prostaglandin by reducing an optically active ethynyl alcoholic derivative.
CONSTITUTION: A compound of formula I (R1 is 1-12C group selected from alkyl which may have alkoxy, cycloalkyl and aralkyl having hetero-atom in aromatic ring or alkyl group; R2 is H or a readily releasable protecting group selected from acyl, silyl, aralkyl and alkyloxyalkyl; * indicates asymmetric carbon atom) is made to react with lithium aluminum hydride, etc., in a solvent such as THF at 40-80°C to reduce the triple bond into trans double bond. Thereby, the objective compound of formula II is obtained. The compound of formula II is used as a starting raw material to simply and efficiently afford an optically active γ-lactone derivative of formula III which is a key intermediate for synthesizing prostaglandin according to the method of G. Stork.
