Abstract:
PURPOSE: To readily produce an osteogenesis-promoting material not containing an osteogenesis-inhibitory substance and, therefor, advantageously applicable to a bone-repairing material or a bone-grafting material in the fields of surgery, orthopaedics and dentistry from a decalcified bone powder in a high yield.
CONSTITUTION: A decalcified bone powder is extracted with a chaotropic reagent and the resultant extract is ultrafiltered to collect a fraction containing a material of 1000-100000 molecular weight. The collected fraction is subsequently dialyzed and a water-insoluble fraction is collected from the dialyzed solution by the conventional method such as centrifugation, thus obtaining an osteogenesis-promoting material not containing an osteogenesis-inhibitory substance in a high yield by using ready processes. In addition, as the raw material bone used for preparation of the decalcified bone powder, a bone of an animal belonging to the same genus as that of the grafted host is preferable. When human is the host, the diaphysis part of the long bone of a mammal such as a rabbit, a pig, a sheep, a horse or a cow, especially a newborn calf is preferably used. The chaotropic reagent is a substance for weakening the bonding between proteins and urea or a guanisine hydrochlorate, especially the latter substance is advantageously used.
