Abstract:

PROBLEM TO BE SOLVED: To obtain the subject compound useful as an intermediate for pharmaceuticals such as paroxetine for antianxiety agent, etc., on an industrial scale in high efficiency by the use of only a small amount of catalyst by introducing a specific glutaric acid alkyl ester into a hydrogen gas atmosphere containing a reduction catalyst.
SOLUTION: The objective compound of formula II [e.g. (±)-cis,trans-4-(4- fluorophenyl)-5-methoxycarbonylpiperidin-2-one] can be produced by introducing (A) a glutaric acid alkyl ester of formula I (R1 and R2 are each a 1-5C alkyl) [e.g. 2-cyano-3-(4-fluorophenyl)glutaric acid dimethyl ester] into a hydrogen gas atmosphere containing (B) a reduction catalyst (e.g. Raney cobalt) preferably under a pressure of atmospheric pressure to about 20 kgf/cm2 at 30-120°C. The components A and B are preferably included or dissolved beforehand in an organic solvent such as methanol.
