Abstract:

PURPOSE: To obtain a recombinant plasmid useful for producing from microbes poly-β-hydroxybutyric acid, a biodegradable thermoplastic resin, without requiring any reduceing substance such as organocarbon source or H2.
CONSTITUTION: This recombinant plasmid pAER1 is obtained by incorporating a vector such as puc19 with a structural gene group coding enzymes composed of β-ketothiolase, acetoacetylCoA reductase and polyhydroxybutyrate polymerase each derived from Alcaligenes eutrophus. This plasmid is transfected into Synechococcus sp. to obtain transformed blue-green algae pAER1-1 (FERM P-14469), which is then cultured to accumulate poly-3b-hydroxybutyric acid in the microbes.
