Abstract:

PURPOSE: To easily obtain the subject compound useful as a synthetic intermediate for cerebral circulation metabolism improver, in high yield, by reacting 3-[2-(dimethylamino)ethoxy]-p-cymene hydrochloride with iodomesitylene diacetate and hydrolyzing the reaction product.

CONSTITUTION: The objective N,N-dimethyl-2-(4-hydroxy-2-isopropyl-5- methylphenoxy)ethylamine of formula III can be produced by reacting a compound of formula I with a compound of formula II in the presence of an acid catalyst (e.g., sulfuric acid or trifluoroacetic acid) in a solvent (e.g., acetic acid) at 0W50°C for 2W48hr, pouring water to the reaction mixture and hydrolyzing the reaction product under refluxing preferably in the presence of a copper salt and an acid. The starting compound of formula I is obtained by reacting thymol with N,N-dimethyl-2-chloroethylamine hydrochloride in the presence of an alkali hydroxide and adding sulfuric acid to the reaction product.
