Abstract:

PROBLEM TO BE SOLVED: To provide a method of determining the interaction interface of protein adjoining other molecules in a complex containing protein with high sensitivity and a high efficiency comparing to conventional methods.
SOLUTION: By substituting deuterium for hydrogen except arbitrary hydrogens (referred to as 'specific hydrogen', below) existing in the side-chains of a protein as one molecule among a plurality of molecules comprising a complex, the position of specific hydrogens existing within 10 &angst from the hydrogens contained in other molecules adjoining the protein is determined based on a cross saturation phenomena using NMR method. As the result, the method enables to determine the interaction interface of the complex containing the protein with high sensitivity and a high efficiency comparing to conventional methods.
