PURPOSE: To purify the titled callicrein for the treatment of cerebrovascular disorder highly, by bringing an aqueous solution containing a crude callicrein derived from a human urine into contact with an insoluble carrier containing specific hydrophobic groups bonded thereto, and using the difference in the affinity.
CONSTITUTION: A crude human callicrein is dissolved in a neutral buffer solution in a suitable salt concentration, and then brought into contact with an insoluble carrier containing hydrophobic groups of the formula (R is ≥4C aliphatic or aromatic hydrocarbon), e.g. agarose or spherically treated agarose. Existing impurities are adsorbed on the carrier by the difference in the hydrophobic nature and separated to give the aimed highly purified callicrein in high yield. The resultant purified callicrein is purified to a purity 10W15 times that of the crude callicrein before the purification, and no colored material is contained therein.
