PURPOSE: To simply, promptly, exactly and inexpensively measure the concentration of microorganism by basing on the observed value of the dissolved oxygen(Do) originating from O2 in H2O2 formed by microorganism catalase in the samples of microorganism suspension.
CONSTITUTION: A sample ot microorganism suspension 15, such as yeast or Escherichia coli in 6-7pH 0.1M phosphate buffer solution is introduced into cell 1, N2 gas is introduced from tube 10 to remove initial DO. Then, H2O2 is introduced from tube 12' into the suspension 15 so that the concentration of H2O2 becomes 10mH (0.01M) at the start of the reaction, and they are stirred with the motor 13 for about 1minute to effect complete mixing to decompose H2O2 with the catalase of the microorganism to increase the 60 concentration originating from O2. The increasing rate of DO is measured with O2 sensor to promptly exhibit the concentration of microorganism on the display or record the value through amplifier 3, A/D converter 5 and microcomputer 7.
