PURPOSE: To prepare a human Cu, Zn-superoxide dismutase gene in high efficiency, by synthesizing a recombinant DNA having a human Cu, Zn-superoxide dismutase structure gene at the downstream of a plasma expression controlling gene.
CONSTITUTION: A microorganism transformed with a recombinant DNA having human Cu, Zn-superoxide dismutase structure gene at the downstream of a plasma expression controlling gene, is cultured, and the human Cu, Zn-superoxide dismutase accumulated in the culture product is separated therefrom. In the case of using Escherichia coli, the cultivation is carried out e.g. in an L medium, an M9 medium containing glucose and casamino acid, etc. usually at about 15W43°C for about 2W24hr. An induced synthesis can be performed by adding a drug such as mitomycin C to the culture product or irradiating the culture product with ultraviolet radiation in logarithmic growth stage.
