Abstract:
PURPOSE: To obtain the constituting molecule containing a DNA encoding a tumor necrosis factor(TNF), having improved cytotoxicity and cytostatic activity against tumor cells, capable of mass producing TNF-like polypeptide such as antitumor agent, etc., in a prokaryotic cell.
CONSTITUTION: A recombinant DNA molecule selected from the group consisting of a DNA insert (DSM 3, 159) of p-mouse tumor necrosis factor (mTNF)-3, a DNA insert (DSM 3, 160) of p-human tumor necrosis factor (hTNF) 1, a DNA sequence hybridizing either or both of these DNA inserts and encoding a polypeptide showing biological activity in expression and a DNA sequence foregoing either of the DNA insert or the DNA sequence, acting on and bonded to an expression control sequence such as T4, pL-T4 or the like is expressed in a prokaryotic host and the prepared TNF-like polypeptide-containing composition is brought into contact with an anion exchanger and the adsorbed material is eluted to give the TNF-like polypeptide having antitumor activity in a high yield.
