Abstract:

PROBLEM TO BE SOLVED: To obtain the subject new toxin fragment having a specific amino acid sequence, not containing castor bean cell, obtained by recombinant genetic engineering, having cytotoxicity and used for medicines or the like for killing target cell such as tumor cell.
SOLUTION: This new recombinant ricin B fragment comprises a recombinant ricin B not containing castor bean cell substance and is subunit of ricin having cytotoxicity and produced by Escherichia coli or yeast cell transformed with DNA encoding an amino acid sequence represented by the formula, and the ricin B subunit has action attaching A subunit having cytotoxicity to a target cell to be killed and promoting transfer of A subunit to the interior of cytoplasm. The ricin B is obtained by screening cDNA library prepared according to ordinary method by using mRNA extracted from seed of Ricinus communis L. and expressing DNA encoding the resultant ricin B with Escherichia coli or yeast cell.
