PURPOSE: To efficiently recover protein existing in a very small amount by passing a protein-containing solution through a rotary column having a built-in carrier for protein adsorption, contacting protein with the carrier, cleaning the column with a cleaning solution and eluting protein adsorbed on the column.
CONSTITUTION: A protein-containing solution (e.g. lactoferrin-containing milk) is passed through a revolving rotary column {length based on volume Vo is L≥(105Vo)1/3; number of revolutions r is 1rpm≤r≤1.000 rpm; velocity of solution passage Vs(l/hr) is Vs(102 Vo; time of solution passage T(hr) is T≤500qV1/CVS [C (g/l) is concentration of protein liquid to be recovered; q is saturated adsorbed amount (g/l gel) of carrier; V1 is volume (l) of built-in carrier} having a built-in carrier (e.g. sulfated polysaccharide) for protein adsorption, brought into contact with the carrier, the column is cleaned with an aqueous solution of sodium chloride and protein adsorbed on the carrier is eluted with the aque ous solution of sodium chloride to recover the aimed protein.
