Abstract:
PURPOSE: To simply and efficiently carry out optical resolution in a large scale by treating a racemic mixture of β-adrenergic propanolamine with a chiral aralkylisocyanate and resolving the resultant diastereomeric urea intermediate and subjecting the resolved compound to hydrazine treatment.
CONSTITUTION: A β-adrenergic aryl- or hetaryl-oxypropanolamine of formula I [Z is an aryl, a hetaryl or the like; Y is a 1-10C alkyl, a group of formula AN (A is a 1-10C alkyl; B is Z, furan, benzofuran or the like); X is H or a group of formula II (R is a 1-10C alkyl, phenyl or the like)] is treated with a chiral isocyanate of formula III (Ar is an aryl; R1 is a 1-6C alkyl) to produce a pair of diastereomeric urea derivatives of urea derivative of formula IV, e.g. N-[3-(2-cyanophenoxy)-2-hydroxypropyl]-N-[1,1-dimethyl-2-(1H-indol-3-yl)- ethyl]-N'-[1,(1-naphthyl)ethyl]urea.
