Abstract:
PURPOSE: To efficiently separate oligomannuronic acids according to polymerization degree by treating a mixture of the oligomannuronic acids different in polymerization degree by the anion-exchange chromatography using a bicarbonate compound solution as an elute.
CONSTITUTION: A mixture of oligomannuronic acids different in polymerization degree is subjected to anion-exchange chromatography treatment using a bicarbonate compound solution as an elute to separate the oligomannuronic acids according to the polymerization degree. Specifically, alginic acid is decomposed in the presence of polyglucuronic acid lyase contained in alginolytic enzymes derived from Flavobacterium-multiboram K-11 (FERM-P-1138) to obtain polymannuronic acid. The resultant polymannuronic acid is subsequently decomposed in the presence of polymannuronic acid lyase derived from an abalone- acetone powder to obtain an oligomannuronic acid mixture. The obtained mixture is then dissolved in an ammonium bicarbonate solution, adsorbed on an anion-exchange resin and subsequently eluted by a linear concentration gradient of an ammonium bicarbonate solution.
