PURPOSE: To provide a shuttle vector, containing a plasmid pMA1 of Microcystis aeruginosa, a multicloning region of a plasmid pUC and an OrieE region of a colicin E1-based plasmid and replicable in Escherichia coli and a unicellular blue-green alga.
CONSTITUTION: This new shuttle vector is obtained by treating (A) a plasmid pMA1 derived from Microcystis aeruginosa with a restriction enzyme HincII, reacting the resultant treated plasmid pMA1 with a plasmid pUC19, derived from Escherichia coli and treated with the restriction enzyme HincII in the presence of a T4 ligase, constructing (B) a shuttle vector pUC19-pMA1, then treating the constructed shuttle vector with a restriction enzyme HindIII, further treating the prepared treated substance with (C) an OriE region of a colicin E1-based plasmid pUC18 in the presence of the T4 ligase and thereby integrating a luciferase gene, etc., therein. The shuttle vector is replicable in Escherichia coli and a unicellular blue-green alga.
