To selectively regulate an extension reaction within a desired area in a substrate.
An oligo probe provided with a caged compound at the terminus thereof is immobilized to a reaction area in a substrate. After pouring a reaction solution into a flow cell including the reaction area, the reaction area alone is irradiated with light. As a result, the photodecomposition-active protecting group at the terminus of the oligo probe that has been immobilized in the reaction area is cleaved off and thus the initiation of a polymerase extension reaction is selectively regulated. In the flow cell, a plural number of reaction areas are located at constant intervals on the substrate. The flow cell fixed to a moving stage is moved by a distance equal to the interval between adjacent reaction areas and then light irradiation is carried out. Next, the extension reaction is measured continuously. By repeating these operations, the base extension reaction can be stably measured in the flow cell without using complicated channels nor replacing the reaction solution.
