Abstract:

To obtain a stable-quality staining composition with high toluidine blue-O proportion, to provide a method for producing the composition, also to provide a method for confirming malformed tissue using the composition, and to provide a method for analyzing the composition.

This method for producing the subject staining composition comprises oxidation of N,N-dimethyl-p-phenylenediamine to form 2-amino-5- dimethylaminophenylthiosulfonic acid followed by its oxidation and then condensation reaction with o-toluidine to form indaminethiosulfonic acid which, in turn, is oxidized to effect cyclization of the indamine ring to produce a reaction product containing toluidine blue-O(TBO); wherein, prior to forming the reaction product, a complexing reagent is introduced into the system.
