Abstract:

PROBLEM TO BE SOLVED: To convert a carbonyl compound into a corresponding optically active alcohol in an excellent stereoselectivity or stereospecificity by reacting a carbonyl compound with a Meerwein.Ponndorf.Verley intermediating substance.
SOLUTION: A compound having a chiral center and a carbonyl group having a prochiral C-atom is brought into contact with an MPV intermediate substance (preferably aluminum sec-butoxide, aluminum isopropoxide, or the like) preferably in the presence of a strong acid (e.g. trifluoroacetic acid) at about ≤50°C for a time sufficient to obtain an excess amount of a desired optically active alcohol to perform a stereoselective reduction for converting into a corresponding optically active alcohol. Preferably, the MPV intermediate substance exists in an amount less than the stoichiometric amount in the reaction and the hydrogen donor is preferably added in at least about 2 times of stoichiometric amount. The method is useful in view of stereoselectivity, stereospecificity and cost.
