Abstract:

PROBLEM TO BE SOLVED: To obtain a new DNA which codes for caludin family of 'tight junction'-constituting membrane protein having a specific amino acid sequence, relates to proliferation and differentiation of cells, inflammation, cancer metastasis, and so on, and gives useful proteins in medical field.
SOLUTION: This is a new DNA which codes for claudin family of 'tight junction'-constituting membrane protein, codes for claudin-1 having an amino acid sequence shown by the formula, is one of intracellular adhesion mechanisms in epithelial and endothelial cell layers, and is useful, for example, for producing membrane proteins useful in medical field. This DNA is obtained by fractionating, using discontinuous sucrose density gradient centrifugation, an adhesion mechanism fraction prepared from chicken liver, analyzing it by SDS- electrophoresis, identifying a protein which is accompanied with occludin, carrying out a homology screening basing on it, followed by screening a mouse cDNA library basing on a sequence of EST clone obtained.
