PURPOSE: To efficiently determine the base sequence of a long DNA.
CONSTITUTION: DNA probe elements holding divided DNA oligomers having various sequences, respectively, and fluorescent substance-labeled oligomer sets produced by labeling the sets of the oligomers with various fluorescent substances, respectively, are employed to simultaneously determine the plural sites of a long DNA or the base sequence of plural DNAs. Primers complementary to the determined sequence are subsequently again selected and used for elongating the length of the determined base sequence. Since the determination of partial sequences, the problem point of a conventional method, and the repeating syntheses of new primers on the basis of the determined sequence can be avoided, the sequence can efficiently be determined without requiring any labor.
