PURPOSE: To breed a new cyclamen having excellent proliferation power by infecting a cyclamen with Agrobacterium, proliferating and subculturing the obtained transformant in a medium free from phytohormone and subjecting to differentiation and induction in a medium containing a phytohormone.
CONSTITUTION: A developed leaf of cyclamen (Cyclamen persicum) is collected, washed, sterilized and thoroughly drained. A circular disk is punched from the leaf with a paper punch to obtain a specimen containing the leaf vein. The specimen is immersed for 10min in a cultured liquid of Agrobacterium obtained by culturing Agrobacterium A4 strain in a culture medium for a night to effect the infection of cyclamen with Agrobacterium. The infected cyclamen specimen is cultured in a medium to induce a transformant, which is subjected to proliferation and subculture in a medium free from phytohormone. The subcultured product is differentiated and induced in a medium containing a phytohormone to obtain the transformed cyclamen.
