PURPOSE: To provide a granular preparation containing 2W80%, based on the weight of the granule, of an osmotic pressure regulator in the granule coated with a coating film, capable of keeping the effective blood concentration of a drug for a long time, stabilizing the drug, and releasing the drug continuously without causing the decomposition and inactivation of the drug with intestinal juice.
CONSTITUTION: The objective granule contains 2W80%, preferably 30W70%, based on the weight of the granule, of an osmotic pressure regulator, and is coated with a coating film. The osmotic pressure regulator is preferably a substance giving high osmotic pressure with a small amount, and giving an environment having low pH in the granule, e.g. an organic acid such as tartaric acid, succinic acid, citric acid, etc., inorganic acid such as boric acid, etc., inorganic salt such as potassium chloride, etc. The granular composition can be prepared by coating a spherical core composed mainly of the osmotic pressure regulator with a coating agent composed of the drug component, a vehicle and optionally the osmotic pressure regulator, and further coating the coated granule with a coating film.
