Abstract:

PURPOSE: To produce a peptide having the activity to promote the action of insulin, by treating a serum albumin of mammal with a specific enzyme.

CONSTITUTION: A mammalian serum albumin is decomposed with one or more enzymes selected from tyrpsin, acrosin, koknase and E. coli protease II to produce a peptide having the activity to promote the action of insulin in the reaction liquid. The treatment with the enzyme is carried out preferably under the conditions shown in the table. The peptide can be separated from the reaction liquid e.g. by molecular filtration with a gel-filtration carrier, dialysis, electrophoresis, ion exchange column chromatography, high performance liquid column chromatography, etc. The separated peptide is purified by dissolving the peptide in a weakly acidic solution, carrying out the gel-filtration and freeze-drying, dissolving in a buffer solution, subjecting to the cation exchange column chromatography, and dialyzing and freeze-drying the product.
