NEW MATERIAL:Plasminogen activator KYM produced by the mutant KYM-A of human rhabdomyosarcoma cell. Appearance, white powder; molecular weight, two bands between 56,000 and 62,000 (nonreduced type) and between 32,000 and 36,000 (reduced type) by SDS-polyacrylamide gel electrophoresis; solubility, 50μg/ ml in water and salt solution, insoluble in ethanol and ether.
USE: Thrombolytic agent. It is a weakly basic glycoprotein having stronger fibrin- bonding capability than urokinase. It is prepared preferably in the form administrable by intravenous injection.
PREPARATION: The cell KYM-I separated from human rhabdomyosarcoma (presented by Dr. Morimasa Sekiguchi, Medical Science Laboratory, Tokyo University) is cultured, and the mutant KYM-A prepared by the repeated separation, selection and culture of the cell is subjected to the suspension culture under agitation to obtain the titled substance on an industrial scale.
