Abstract:

PURPOSE: To provide an E-coil polycopying plasmid, by introducing a specific fragment to the restriction enzyme site of a starting plasmid.

CONSTITUTION: A gene containing E-coli tryptophan operon in digested with restriction enzyme Bgl II to obtain tryptophan promoter and a DNA fragment containing an operator, a leader region and a part of trpE. A restriction enzyme EcoRI site is formed to the terminal of the fragment, and the fragment is introduced to the restriction enzyme EcoRI site of the E-coli polycopying plasmid. A DNA fragment containing a gene coding β-endorphin is introduced to the EcoRI site of the plasmid.
